摘  要:本试验以黑砖茶为原料,从中分离出灰绿色真菌,通过对其菌落形态观察,和对其孢子进行显微观察以及电镜观察,并通过PCR反应扩增其ITS序列进行比对,以此为基础建立系统发育树,因而从细胞和分子水平上鉴定提取出的灰绿色真菌为灰绿曲霉。将获得的原始灰绿曲霉进行紫外诱变,对比诱变后的菌株与原始菌株在菌落形态和细胞形态上都有差异。两者ITS序列中第67号碱基、255号碱基、387号碱基、388号碱基、521号碱基五处碱基不同,其他碱基序列都相同,以此证明成功的筛选出了灰绿曲霉的突变株。

Screening of Mutants of Aspergilling Glaucus
Abstract: Based on raw materials of black brick tea, the experiment aimed at identifying Aspergillus glaucus isolated at the cellular and molecular level by building a phylogenetic tree basing on observation on colony morphosis, microscopic and sub-microscopic observation on spores in combination with amplifying corresponding ITS sequence via PCR. There has been distinction in both colonial and cellular morphosis between the UV-irratdiated strain and the original strain isolated. ITS sequence of both the strains proved to be fully homologous except 67bp, 255bp, 387bp, 388bp and 521bp, which underlying the conclusion that a mutant of Aspergillus glaucus has been sucssesfully isolated after screening .
