摘  要:本论文旨在研究赤霉素对苎麻的茎皮和顶芽的影响。先分析筛选得到的差异表达基因的数据信息。再在NCBI网站用BLAST软件进行序列比对等生物信息学搜索,并设计检测基因表达水平的RT-PCR引物,根据具体的差异表达序列的特征,设计特异性基因表达PCR引物。利用提取得到的赤霉素处理与对照材料的总RNA。反转录得cDNA,PCR鉴定特异基因在两种材料中的表达水平。通过RT-PCR检测赤霉素对苎麻的茎皮和顶芽有上调作用。

Gene expression regulation by GA treating in the Ramie
Abstract: This paper aims to study the stem bark of ramie gibberellin and crown. The first analysis of screening differentially expressed genes of data. BLAST site in NCBI with software sequence alignment, etc, and the design of bioinformatics search gene expression level detection of primers, RT - PCR according to specific features of the sequence of differentially expressed genes expression design specific PCR primers. Using the extraction of ga3 treatment and contrast material will RNA. Reverse transcriptase have identified genes, PCR cDNA in the expression level of two kinds of material. Through the RT - PCR kowning GA3 has raised ramie of gibberellin stem bark and crown.

本研究是通过设计特异性序列的引物,利用RT-PCR的方法,成功的鉴定了赤霉素对苎麻影响。GA处理的苎麻昆池一号。除了在茎皮里对于Uigene6997_ ramie这个上调基因没有表现出明显的上调作用外,Unigene45141_ ramie和 Unigene1570_ ramie 的上调基因在处理了的苎麻的茎皮和顶芽中都表达出了上调作用。
