观察PCV2的病理变化及病毒的免疫组化分析(9000字)
摘 要：目的：为了研究圆环病毒在侵入动物机体内，在被感染动物体内不同组织器官在不同时间病毒分布情况，采用免疫组化的方法在显微镜下观察被感染动物器官病变情况。实验方法：选择刚断奶的清洁级昆明小鼠28只，分为两组：一组为实验组，共21只，腹腔接种PCV-2病毒液0.2ml；一组为对照组，7只，腹腔注射生理盐水0.2ml。将两组昆明小鼠分别饲养于环境条件相同的不同房间内，给予相同的饲料和饮水。7天后，以剪断颈椎的形式将昆明小鼠处死实验组老鼠3只，对照组1只，观察病理变化；14天后再以相同的处死方法处死每组中的4只昆明小鼠，观察病理变化；以此类推，以后每隔7天就如此处死小鼠，直到将所有的昆明小鼠全部处死。然后做石蜡切片，并做免疫组织化学实验检测病毒。结果：圆环病毒能在短时间内侵入动物各主要器官，且在淋巴结、肝部、脾脏病变迅速且明显。结论：PCV2确实能造成小鼠组织器官病变，免疫组化方法可以作为圆环病毒实验室诊断方法。
